Overview

MNRP1685A + bevacizumab is an investigational combination therapy consisting of MNRP1685A (also known as Vesencumab), a fully human monoclonal antibody targeting neuropilin-1 (NRP1), and bevacizumab, a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A). MNRP1685A blocks the binding of VEGF, VEGF-B, and placental growth factor 2 to NRP1, keeping vessels immature and highly VEGF-dependent, thereby disrupting tumor vasculature maturation and angiogenesis. The rationale for the combination is to enhance anti-angiogenic effects beyond those achieved by targeting VEGF-A alone. This combination was studied for the treatment of advanced or metastatic solid tumors. Clinical trials demonstrated safety profiles consistent with the individual agents, though a higher than expected incidence of clinically significant proteinuria was observed, limiting further development of the combination[1][3][4][6][8].
