modakafusp alfa + pomalidomide + bortezomib is an experimental combination therapy studied for the treatment of relapsed/refractory multiple myeloma. - **Modakafusp alfa** is a novel engineered fusion protein combining Type I interferon (IFN) with an IgG4 backbone, targeting CD38 to deliver IFN signaling directly to malignant plasma cells and modulate immune responses. - **Pomalidomide** is an immunomodulatory imide drug (IMiD) that exerts anti-myeloma activity through modulation of the immune system and inhibition of cell proliferation, notably enhancing T-cell and natural killer (NK) cell-mediated cytotoxicity. - **Bortezomib** is a reversible proteasome inhibitor that induces apoptosis in multiple myeloma cells by disrupting proteasome-mediated protein degradation, leading to accumulation of toxic proteins and cell death. This triplet therapy is being evaluated in clinical trials for efficacy and safety in adult patients with relapsed/refractory multiple myeloma who have previously received at least one line of therapy.
