ModraDoc003 + ModraDoc004

ModraDoc003 and ModraDoc004 are oral tablet formulations of docetaxel developed by the Netherlands Cancer Institute in collaboration with Slotervaart Hospital. Docetaxel is a taxane-based chemotherapy agent that works by stabilizing microtubules, thereby inhibiting cell division and inducing apoptosis in cancer cells. Because docetaxel has poor oral bioavailability due to high first-pass metabolism by the CYP3A4 enzyme and efflux by P-glycoprotein in the gastrointestinal tract, these ModraDoc formulations are co-administered with ritonavir. Ritonavir acts as a pharmacokinetic enhancer (booster) by inhibiting CYP3A4, allowing for therapeutic systemic exposure of docetaxel to be achieved via oral administration. This approach aims to improve patient convenience and quality of life compared to traditional intravenous docetaxel infusions for the treatment of advanced solid tumors.
