Overview

Mogamulizumab + brentuximab vedotin is an investigational combination therapy of two monoclonal antibody-based drugs used in clinical studies for cutaneous T-cell lymphomas, such as mycosis fungoides and Sézary syndrome. **Mogamulizumab** is a monoclonal antibody targeting CC chemokine receptor 4 (CCR4), primarily expressed on malignant T cells; it acts as a CCR4 antagonist, blocking T cell proliferation and chemotaxis, and may also induce antibody-dependent cell-mediated cytotoxicity. **Brentuximab vedotin** is an antibody-drug conjugate composed of an anti-CD30 chimeric antibody linked to the cytotoxic agent monomethyl auristatin E (MMAE) via a protease-susceptible linker; it binds to CD30-expressing cells, is internalized, and releases MMAE to disrupt the microtubule network and cause apoptosis. The combination targets two distinct pathways—the immune microenvironment (CCR4) and the tumor marker (CD30)—for potentially synergistic antitumor activity. Clinical investigation focuses on safety, tolerability, and establishing a safe dosing regimen.
