Overview

Mogamulizumab + tremelimumab is an investigational combination of two immunotherapy monoclonal antibodies. **Mogamulizumab** is an anti-CCR4 humanized monoclonal antibody that targets CC chemokine receptor type 4 (CCR4) on certain regulatory T cells (Tregs) and some malignant T cells, leading to their depletion via mechanisms such as antibody-dependent cell-mediated cytotoxicity (ADCC). **Tremelimumab** is a fully human monoclonal antibody against CTLA-4, blocking the interaction of CTLA-4 with CD80/CD86, which enhances T-cell mediated immune responses. The combination has been evaluated in phase I/early trials in advanced solid tumors—most notably in advanced pancreatic cancer. The main goal was to achieve improved antitumor activity by both depleting Tregs (through CCR4 blockade) and enhancing effector T-cell activation (through CTLA-4 inhibition). Clinical studies to date suggest that the combination is tolerable, with immunity modulation effects seen, but antitumor efficacy was limited in the tested patient populations[1][3][8][5].
