Overview

Mogamulizumab + utomilumab is an investigational combination therapy consisting of two monoclonal antibodies. Mogamulizumab is a humanized monoclonal antibody targeting C-C chemokine receptor 4 (CCR4), which is highly expressed on regulatory T cells (Tregs) and certain tumor cells; it depletes CCR4-positive Tregs, thereby reducing immunosuppression in the tumor microenvironment. Utomilumab (PF-05082566) is a fully human monoclonal antibody agonist of the T-cell costimulatory receptor 4-1BB (CD137), which enhances effector T cell function and promotes antitumor immunity by stimulating cytokine secretion and proliferation of cytotoxic lymphocytes. The combination aims to synergistically enhance antitumor immune responses by both expanding effector T cells and depleting immunosuppressive regulatory T cells. This regimen has been evaluated in phase Ib clinical trials for patients with advanced solid tumors, demonstrating safety, tolerability, pharmacodynamic activity including peripheral depletion of Tregs, and modest clinical activity[1][2][9].
