Overview

MOMA-313 + olaparib is a combination therapy consisting of MOMA-313, a first-in-class oral inhibitor of DNA polymerase theta (Polθ), and olaparib, a well-established oral small molecule inhibitor of poly (ADP-ribose) polymerase (PARP). This combination is being evaluated for the treatment of advanced or metastatic solid tumors characterized by homologous recombination deficiency (HRD), including but not limited to subtypes with BRCA1/2, ATM, PALB2, and other HR-related gene mutations. Both drugs are given orally. MOMA-313 targets tumor cells reliant on alternative DNA repair mechanisms, while olaparib impairs single-strand DNA repair by inhibiting PARP, collectively leading to increased DNA damage and cancer cell death in HRD tumors. This combination is in early clinical investigation and is not yet approved for any indication[1][3][5][7][9].
