The monoclonal antibody 17-1A F(ab')2 fragment is a bivalent antigen-binding fragment derived from the murine monoclonal antibody edrecolomab (also known as CO17-1A or anti-EpCAM/17-1A). The parent antibody targets the epithelial cell adhesion molecule (EpCAM), which is expressed on normal epithelial tissues and a wide range of carcinomas. The F(ab')2 format is generated by enzymatic cleavage (typically with pepsin) of the full-length IgG, removing the Fc region while retaining both antigen-binding arms connected via the hinge region[6][7][1]. This modification reduces Fc-mediated effector functions and immunogenicity but preserves high avidity for EpCAM. The primary mechanism of action involves binding to EpCAM on tumor cells, potentially blocking cell adhesion and marking cells for immune recognition. Historically, edrecolomab was investigated as adjuvant therapy in colorectal cancer; its fragments have been explored for diagnostic imaging and intraoperative radioimmunodetection due to their improved tissue penetration compared to whole antibodies[9][4].
