Overview

Motixafortide is a cyclic peptide and potent inhibitor of the C-X-C motif chemokine receptor 4 (CXCR4). It is used in combination with a granulocyte-colony stimulating factor (G-CSF), such as filgrastim, to mobilize hematopoietic stem cells (HSCs) from the bone marrow into the peripheral blood for collection and subsequent autologous transplantation in patients with multiple myeloma. Motixafortide blocks CXCR4, preventing its interaction with its ligand CXCL12 (also known as stromal-derived factor 1α or SDF-1α), which normally helps retain HSCs within the bone marrow. By inhibiting this pathway, more stem cells are released into circulation, improving collection rates for transplantation. The combination has demonstrated superiority over G-CSF alone in clinical trials for mobilizing sufficient numbers of primitive HSCs for transplant. Motixafortide is marketed under the brand name Aphexda and was developed by BioLineRx[1][3][4][5][6][7].
