**MP0250 + osimertinib** is an investigational drug combination for the treatment of **EGFR-mutated, T790M-positive non-small cell lung cancer (NSCLC)**. **MP0250** is a multi-specific DARPin (Designed Ankyrin Repeat Protein) fusion protein that simultaneously binds and neutralizes both vascular endothelial growth factor A (VEGF-A) and hepatocyte growth factor (HGF), thereby targeting tumor angiogenesis and interfering with microenvironment-mediated mechanisms of drug resistance. **Osimertinib** is a third-generation, irreversible small molecule inhibitor targeting epidermal growth factor receptor (EGFR) mutations, including T790M, a key resistance mutation in NSCLC. The rationale for the combination is that while osimertinib continuously inhibits the EGFR pathway, MP0250 blocks key resistance escape pathways (VEGF and HGF/cMET), aiming to overcome acquired resistance in treated NSCLC. This combination has been tested in early-stage clinical trials by Molecular Partners AG.
