mproUK + low-dose tPA is an intravenous dual thrombolytic regimen developed by Thrombolytic Science International (TSI) for the treatment of ischemic stroke and acute myocardial infarction. The therapy consists of a mutant form of pro-urokinase (mproUK, also known as HisproUK or M5) and a low dose of alteplase (tPA). This combination is designed to mimic the body's natural physiological fibrinolytic mechanism: a small dose of tPA initiates clot lysis by activating fibrin-associated plasminogen, which then creates a binding site for mproUK to complete the dissolution of the fibrin clot. This sequential action aims to provide more efficient and safer thrombolysis compared to standard-dose tPA monotherapy, specifically by reducing the risk of symptomatic intracranial hemorrhage and preserving plasma fibrinogen levels.
