MPTP + probenecid refers to an experimental drug combination used primarily to create a chronic and progressive animal model of Parkinson’s disease, most notably in mice. **MPTP** (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) is a neurotoxin that selectively destroys dopaminergic neurons in the substantia nigra by being metabolized to MPP+, which inhibits mitochondrial complex I, leading to dopaminergic cell death. **Probenecid** is a uricosuric agent and renal tubular secretion inhibitor; when co-administered, it impairs the renal clearance of MPTP and its active metabolites, thereby prolonging and intensifying MPTP’s neurotoxic effects and promoting chronic neurodegeneration. This combination results in a progressive dopamine neuron loss and behavioral deficits that more faithfully recapitulate key features of human Parkinson’s disease compared to acute MPTP treatment[1][2][5]. The model is widely used in PD research to evaluate experimental neuroprotective therapies and study pathophysiology.
