MRG003 + pucotenlimab

MRG003 + pucotenlimab is a combination therapy under clinical investigation for the treatment of solid tumors, particularly squamous cell carcinoma of the head and neck (SCCHN) and nasopharyngeal carcinoma (NPC). MRG003 is an antibody-drug conjugate (ADC) targeting epidermal growth factor receptor (EGFR), designed to deliver cytotoxic agents directly to EGFR-expressing tumor cells. Pucotenlimab, also known as HX008 or Puyouheng, is a recombinant humanized IgG4 monoclonal antibody that inhibits programmed cell death protein 1 (PD-1), thereby blocking its interaction with PD-L1 and PD-L2 to restore immune-mediated tumor cell killing. Preclinical studies have shown synergistic antitumor effects when these two agents are combined. Clinical trials have demonstrated good tolerability and encouraging antitumor activity in patients with locally advanced or metastatic EGFR-positive solid tumors, especially in those who failed prior PD-1 therapy[1][3][5].
