Overview

**mRNA-5671 + V176** is a combination therapy consisting of mRNA-5671 (also known as V941), an investigational lipid nanoparticle-formulated mRNA cancer vaccine developed by Moderna that encodes four common mutant KRAS proteins (G12C, G12D, G12V, G13D) to stimulate T-cell responses against KRAS-mutated tumors, paired with V176, an investigational PD-1 inhibitor monoclonal antibody from Merck that blocks the PD-1 receptor to enhance antitumor immunity. The vaccine is designed as an off-the-shelf immunotherapy for KRAS-driven cancers, inducing mutant-specific CD8+ T cells and IFN-γ production as measured by ELISpot assays, while V176 potentiates T-cell activity by inhibiting immune checkpoints. Primarily evaluated in phase 1 trials for advanced solid tumors with KRAS mutations, including non-small cell lung cancer (NSCLC), colorectal cancer, and pancreatic adenocarcinoma, often in combination with other agents like pembrolizumab in related studies, though specific data on this exact pairing is limited.[1][2][3][9][15]
