Overview

**mRNA-5671 + V940** is a combination therapy consisting of mRNA-5671 (also known as V941), an investigational mRNA-based cancer vaccine from Moderna that encodes four prevalent mutant KRAS antigens (G12D, G12V, G13D, G12C) to stimulate T-cell responses against KRAS-mutant tumors, administered in combination with V940 (mRNA-4157), Moderna's individualized neoantigen therapy (INT) mRNA vaccine encoding patient-specific tumor neoantigens. Developed through a collaboration between Moderna and Merck, this regimen is designed for advanced solid tumors harboring KRAS mutations, such as non-small cell lung cancer (NSCLC), colorectal cancer, and pancreatic adenocarcinoma, with administration via intramuscular injection for mRNA-5671/V941 and intravenous pembrolizumab in trial arms. It aims to induce robust, targeted anti-tumor immunity by combining shared KRAS targeting with personalized neoantigen specificity, evaluated primarily for safety, tolerability, and preliminary efficacy in Phase 1 trials.[1][2][3][4][5]
