Drug intelligence / Profile preview
mRNA-engineered dendritic cell vaccine (TetraMix) is an autologous, personalized, monocyte-derived dendritic cell (moDC) cancer vaccine. The vaccine is engineered ex vivo by electroporating patient-derived moDCs with patient-specific tumor neoantigen mRNA along with TetraMix, a novel four-component mRNA-based adjuvant. TetraMix consists of four mRNA molecules encoding constitutively active toll-like receptor 4 (caTLR4), CD40 ligand (CD40L), interferon-gamma (IFN-gamma), and a decoy receptor for interleukin-10 (dIL10Ralpha). This combination of adjuvant mRNAs redirects the dendritic cells into an antiviral-like transcriptional program, enhancing their maturation, increasing interleukin-12 (IL-12) production, and promoting the activation of tumor-specific cytotoxic CD8+ T cells. Originally developed by the Vrije Universiteit Brussel (VUB) and licensed to etherna (formerly eTheRNA immunotherapies), the vaccine was also positioned for clinical development in colorectal cancer by the Persomed consortium, though clinical trials were halted due to funding constraints.
