mSAR'877 is a murine surrogate of SAR445877, a novel antibody-cytokine fusion protein developed by Sanofi. It consists of a murine anti-PD-1 antibody fused to a mutated murine interleukin-15 (IL-15) and IL-15 receptor alpha sushi domain. The molecule is designed to cis-activate effector T cells by biasing IL-15 stimulation specifically to T cells expressing PD-1, thereby enhancing anti-tumor activity while potentially reducing systemic toxicities associated with untargeted IL-15. In preclinical B16-OVA mouse models, mSAR'877 has demonstrated superior efficacy compared to anti-PD-1 monotherapy or untargeted IL-15, characterized by increased tumor immune cell infiltration (CD8+ T cells and NK cells) and tumor regression.
