MSC-Cytokine + anti-PD-L1 is an investigational combination cell therapy being developed by Kyttaro Therapeutics for the treatment of various cancers. The therapy utilizes Kyttaro's MSC-Cyt platform, which consists of mesenchymal stromal cells (MSCs) genetically engineered to secrete the immunostimulatory cytokines interleukin-7 (IL-7) and interleukin-12 (IL-12). This cell-based delivery system is used in conjunction with a PD-L1 checkpoint inhibitor. The mechanism of action involves the modified MSCs homing to the tumor microenvironment and locally releasing IL-7 and IL-12 to promote T-cell survival, proliferation, and activation. Simultaneously, the PD-L1 blockade prevents the tumor from suppressing the immune response, thereby enhancing the overall anti-tumor efficacy. The therapy is currently in the research and development stage, with potential applications in glioblastoma, colon cancer, melanoma, and metastatic disease.
