MSLN CAR-T (cRNA) refers to a type of Chimeric Antigen Receptor (CAR) T-cell therapy designed to target Mesothelin (MSLN), a tumor-associated antigen highly expressed in various solid tumors such as mesothelioma, pancreatic, ovarian, and lung cancers. In this approach, T cells are engineered to express a CAR that specifically recognizes MSLN on cancer cells. The "cRNA" (circular RNA, often used interchangeably with mRNA in this context) component indicates that the CAR gene is delivered via messenger RNA, leading to transient expression of the CAR on the T-cell surface. This transient expression is a strategy employed to potentially enhance the safety profile of the therapy by reducing the risk of prolonged on-target/off-tumor toxicity, a significant challenge for CAR-T therapies in solid tumors. Upon binding to MSLN, the engineered T cells are activated to induce an anti-tumor immune response and kill the cancer cells.
