MT-101 + fludarabine + cyclophosphamide is a combination regimen consisting of **MT-101**, an autologous mRNA-engineered chimeric antigen receptor (CAR) monocyte cell therapy targeting the CD5 surface receptor, and the lymphodepletion agents **fludarabine** (a nucleoside analog) and **cyclophosphamide** (an alkylating agent). MT-101 is designed to modify the tumor microenvironment, promote anti-tumor immunity, and specifically recognize and kill CD5+ cancer cells. The combination employs fludarabine and cyclophosphamide as lymphodepleting chemotherapy agents to reduce the patient's endogenous T cells, enhancing engraftment and function of the infused CAR monocytes[1][5][7]. MT-101 is currently developed and investigated by Myeloid Therapeutics for relapsed/refractory peripheral T-cell lymphoma (PTCL), especially CD5+ subtypes, in an ongoing phase 1/2 clinical trial.[1][5][7][9]
