MT-8421 + nivolumab is an investigational combination therapy for advanced solid tumors, currently under early-phase clinical development. MT-8421 is a novel engineered toxin body targeting CTLA-4, designed to selectively deplete regulatory T cells (Tregs) within the tumor microenvironment through ribosome inactivation, thereby enhancing antitumor immune responses. Nivolumab is a monoclonal antibody checkpoint inhibitor targeting the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1/PD-L2 to promote T cell-mediated antitumor activity. The combination aims to produce synergistic immune activation by simultaneously reducing immune suppression (via Treg depletion) and augmenting T cell function (via PD-1 blockade).
