Overview

MT1110 + LRRK2 inhibitor is a preclinical-stage small molecule combination therapy being developed by Montara (also referred to as Montaur Life Sciences) for the treatment of Parkinson's disease. The therapy utilizes Montara's proprietary BrainOnly platform, which pairs a brain-penetrant, target-specific drug (a Brain-Targeting Chimera or BrainTAC) with a non-brain-penetrant 'universal' peripheral blocker, MT1110. MT1110 works by saturating FKBP12 (FK506-binding protein 12) binding sites in peripheral tissues, preventing the LRRK2 inhibitor—which is engineered to be FKBP12-dependent—from engaging its target outside the brain. This brain-selective pharmacology aims to avoid the dose-limiting peripheral toxicities (such as lung abnormalities) typically associated with systemic LRRK2 inhibition, thereby expanding the therapeutic index for Parkinson's disease patients.
