MTBhsp70-scFv(MSLN) fusion protein is a recombinant biologic immunotherapy designed to target mesothelin-expressing tumors while simultaneously providing a potent immune-activating adjuvant. It consists of a single-chain variable fragment (scFv) specific for mesothelin (MSLN) linked to the Mycobacterium tuberculosis heat shock protein 70 (MTBHsp70). The scFv component directs the protein to MSLN-overexpressing cells, such as those in ovarian cancer and mesothelioma, while the MTBHsp70 component acts as an adjuvant by binding to CD40 on antigen-presenting cells (APCs). This interaction triggers dendritic cell activation, enhances antigen cross-presentation, and stimulates robust CD8+ T-cell and NK-cell mediated anti-tumor responses. Developed primarily in academic settings, it has demonstrated efficacy in preclinical murine models by delaying tumor onset and improving survival.
