Drug intelligence / Profile preview
MTX-LDE is an experimental therapeutic formulation consisting of the anti-inflammatory and antineoplastic agent methotrexate (MTX) incorporated into lipid core nanoparticles (LDE). Developed by researchers at the University of Sao Paulo, the LDE delivery system is designed to mimic the structure and composition of low-density lipoproteins (LDL). This biomimetic approach allows the nanoparticles to be recognized by LDL receptors, which are often overexpressed in tissues experiencing intense inflammation or rapid cell proliferation. In the context of COVID-19, MTX-LDE is being evaluated for its ability to target inflamed lung tissue and systemic inflammatory cells to mitigate the hyperinflammatory state, or 'cytokine storm,' associated with the disease. By concentrating the drug at the site of inflammation, the formulation aims to enhance the therapeutic index of methotrexate while minimizing systemic toxicities typically associated with conventional methotrexate administration.
