MUC1-KLH + QS-21 is an investigational therapeutic cancer vaccine developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of epithelial malignancies, specifically ovarian, fallopian tube, and primary peritoneal cavity cancers. The vaccine targets the MUC1 glycoprotein, which is frequently overexpressed and aberrantly glycosylated on the surface of various cancer cells. It consists of a synthetic, glycosylated MUC1 peptide conjugated to the immunogenic carrier protein keyhole limpet hemocyanin (KLH) to enhance immune recognition. The formulation is administered with the saponin-based adjuvant QS-21 to further stimulate a robust immune response, primarily aimed at inducing high-titer IgM and IgG antibodies against MUC1-expressing tumor cells. Clinical development has focused on Phase I trials evaluating the safety, optimal dosage, and immunogenicity of this monovalent construct in patients with recurrent or refractory disease.
