Muromonab-CD3 + cyclophosphamide is a combination therapy investigated for the treatment of metastatic cancer and locally progressive primary brain tumors. Muromonab-CD3 (OKT3) is a murine monoclonal antibody that targets the CD3 epsilon chain of the T-cell receptor complex, leading to T-cell depletion and immunosuppression. In this clinical trial context, it was combined with low-dose cyclophosphamide, an alkylating agent that, at low doses, is thought to enhance anti-tumor immune responses by selectively depleting regulatory T cells (Tregs). The study was sponsored by St. Vincent Medical Center - Los Angeles to evaluate the safety and efficacy of this immunomodulatory approach in advanced malignancies.
