Drug intelligence / Profile preview
Muzastotug + atezolizumab + bevacizumab is an investigational triplet immunotherapy regimen combining muzastotug (ADG126), a masked, tumor-activated anti-CTLA-4 monoclonal antibody, with atezolizumab, an anti-PD-L1 monoclonal antibody, and bevacizumab, an anti-VEGF monoclonal antibody. Muzastotug is designed using Adagene's SAFEbody technology to conditionally activate in the tumor microenvironment, minimizing systemic toxicities while selectively depleting regulatory T cells (Tregs) and blocking CTLA-4 signaling. Atezolizumab blocks PD-L1 to restore T-cell anti-tumor activity, and bevacizumab inhibits VEGF-A to suppress angiogenesis and alleviate immunosuppression in the tumor microenvironment. This triplet combination is being developed under a clinical collaboration between Adagene and Roche, and is currently being evaluated in the Phase 1b/2 MORPHEUS-Liver umbrella study (NCT04524871) as a first-line treatment for patients with locally advanced, metastatic, or unresectable hepatocellular carcinoma (HCC).
