Overview

MVA-BN-brachyury + FPV-brachyury is a novel prime–boost therapeutic cancer immunotherapy regimen. It consists of a priming vaccine (MVA-BN-brachyury, also known as MVA-BN-brachyury-TRICOM), which uses a recombinant modified vaccinia Ankara viral vector encoding the human transcription factor and tumor-associated antigen brachyury, along with three T-cell co-stimulatory molecules (B7-1, ICAM-1, LFA-3; collectively called TRICOM). This is followed by multiple booster doses of fowlpox virus (FPV)-brachyury vaccine encoding the same antigens. The therapy is designed to induce a cytotoxic T-cell immune response specifically against tumor cells expressing brachyury—a transcription factor overexpressed in many advanced solid tumors and associated with metastasis and treatment resistance. The regimen has been developed primarily for metastatic or unresectable malignant solid tumors such as chordoma and other sarcomas[1][2][3][4][5][6][8].
