MVT-5873 + gemcitabine + nab-paclitaxel is an investigational combination regimen composed of MVT-5873, a fully human IgG1 monoclonal antibody targeting carbohydrate antigen 19-9 (CA 19-9, also known as Sialyl Lewis a), with two cytotoxic chemotherapeutic agents: gemcitabine (a nucleoside analog inhibiting DNA synthesis) and nab-paclitaxel (paclitaxel formulated as albumin-bound nanoparticles). MVT-5873 induces cancer cell death via antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC), offering targeted immune-mediated antitumor activity in CA 19-9 positive malignancies, especially pancreatic cancer[1][2][3][4][5][6][7]. Gemcitabine and nab-paclitaxel together are a standard backbone therapy for metastatic pancreatic adenocarcinoma, acting through DNA synthesis inhibition and microtubule stabilization, respectively. The combination is developed and investigated primarily in advanced and operable pancreatic, biliary, and colorectal cancers showing CA 19-9 elevation.
