Overview

N,N-dimethylglycine (DMG) is a naturally occurring amino acid derivative found in the cells of all plants and animals and can be obtained in small amounts from dietary sources such as grains and meat. It is produced endogenously during the metabolism of choline to glycine. DMG has been widely used as a dietary supplement for purported benefits including improved physical performance and endurance; however clinical studies have shown little to no effect for these indications. Mechanistically, DMG may act at the glycine binding site of the NMDA receptor and has been proposed to enhance oxygen utilization by tissues and function as an antioxidant by serving as a source of glycine for glutathione synthesis. It also exhibits anti-inflammatory effects in cellular models. Despite its use in various conditions such as ADHD, epilepsy, chronic fatigue syndrome (CFS), allergies, respiratory disorders and multiple sclerosis among others—there is insufficient evidence supporting its efficacy for these uses[1][3][4][5][6][8].
