Drug intelligence / Profile preview
N-(4-chlorophenyl)-5-nitro-3-thiophenecarboxamide is a novel, low molecular weight, narrow-spectrum small molecule antimicrobial candidate under preclinical development for the treatment of Helicobacter pylori infections. Developed by the University of Zaragoza and the Institute for Health Research Aragón (IIS Aragón), the compound acts as a potent inhibitor of the essential OmpR-like orphan response regulator HsrA (HP1043), which is unique and highly conserved among Epsilonproteobacteria (Campylobacterota) and crucial for bacterial cell viability. By binding to HsrA, the compound inhibits its DNA-binding activity, disrupting the transcription of essential genes involved in transcription, translation, redox homeostasis, and energy metabolism. In preclinical mouse models, oral administration of this compound significantly diminished gastric colonization of H. pylori, demonstrating its potential as a novel therapeutic option to overcome multidrug-resistant strains without causing widespread dysbiosis of the host microbiota.
