n-[3-(2,5-dimethoxybenzyloxy)-4-(methanesulfonyl-methylamino)phenyl]-4-bromobenzamide is a synthetic small molecule and a derivative of the nonsteroidal anti-inflammatory drug (NSAID) nimesulide. It is designed to function as a tubulin polymerization inhibitor, targeting the tubulin protein which is essential for cell division, shape maintenance, and intracellular transport. By binding to tubulin, the compound disrupts microtubule dynamics, leading to cell cycle arrest and apoptosis, particularly in malignant cells that exhibit high tubulin activity. Preclinical studies have demonstrated that this compound possesses potent antiproliferative activity against breast cancer (MDA-MB-468) and prostate cancer (DU145) cell lines, with IC50 values of 3.89 nM and 2.30 nM, respectively. The molecule's structure includes a 2,5-dimethoxybenzyloxy group and a 4-bromobenzamide moiety attached to a modified nimesulide scaffold.
