N-{(4S)-4-Amino-5-[(2-aminoethyl)amino]pentyl}-N'-nitroguanidine is a synthetic small molecule that acts as a highly selective inhibitor of neuronal nitric oxide synthase (nNOS). It is used primarily in biochemical and pharmacological research to study the role of nitric oxide in neural signaling, vascular tone regulation, immune function, and cellular signaling pathways. The compound demonstrates strong selectivity for nNOS over other isoforms such as endothelial (eNOS) and inducible (iNOS) nitric oxide synthases, with reported Ki values around 120 nM and >2500-fold selectivity for nNOS versus eNOS. Its mechanism involves binding to the heme domain of nNOS, thereby inhibiting enzymatic production of nitric oxide in neurons[6][7][9]. This property makes it valuable for investigating neurodegenerative disorders and other conditions where modulation of neuronal NO production is relevant.
