Overview

A combination of n-3 polyunsaturated fatty acids (PUFAs), commonly known as omega-3 fatty acids (including α-linolenic acid [ALA], eicosapentaenoic acid [EPA], and docosahexaenoic acid [DHA]), and conjugated linoleic acid (CLA), a group of positional and geometric isomers of linoleic acid. This combination is used primarily as a dietary supplement for its potential anti-inflammatory, immunomodulatory, metabolic, and body composition effects. Mechanistically, both components modulate peroxisome proliferator-activated receptors (PPARs): CLA predominantly activates PPAR gamma to exert anti-inflammatory effects and regulate adipogenesis; n-3 PUFAs activate PPAR delta and suppress NFκB signaling to reduce inflammation. The combination has been studied in animal models for inflammatory bowel disease (IBD) where it showed synergistic upregulation of keratinocyte growth factor but mixed results on disease onset and recovery[1][2]. In humans, supplementation may help prevent abdominal fat mass accrual in younger obese individuals without adversely affecting insulin sensitivity[4]. Both ingredients are also associated with improved markers of inflammation and oxidative stress in cardiovascular disease[8]. However, most evidence comes from preclinical or small clinical studies; large-scale clinical efficacy remains unproven.
