N-803 (also known as ALT-803) is a novel immunotherapeutic agent classified as an interleukin-15 (IL-15) superagonist and antibody-cytokine fusion protein. It stimulates the proliferation and activation of natural killer (NK) cells and CD8+ T cells, thereby enhancing anti-tumor immune responses. Atezolizumab is a monoclonal antibody that targets programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 to prevent immune suppression by tumor cells. The combination of N-803 with atezolizumab aims to synergistically boost the immune system's ability to attack cancer by both activating effector immune cells and relieving checkpoint inhibition. This combination is under investigation for various cancers, including non-small cell lung cancer (NSCLC), triple-negative breast cancer (TNBC), Merkel cell carcinoma (MCC), and non-muscle invasive bladder cancer[1][3][5].
