N-803 + broadly neutralizing antibodies is an investigational combination therapy being studied primarily for HIV infection. N-803 is a novel interleukin 15 (IL-15) superagonist complex consisting of an IL-15 mutant (IL‑15N72D) bound to an IL‑15 receptor α/IgG1 Fc fusion protein. It directly stimulates CD8+ T cells and natural killer (NK) cells through beta gamma T-cell receptor binding, generating memory T-cells while avoiding regulatory T cell stimulation. Broadly neutralizing antibodies (bNAbs), such as VRC07‑523LS and 10‑1074, are monoclonal antibodies that target conserved regions on the HIV envelope glycoprotein and can block viral entry into host cells as well as mediate immune clearance of infected cells. The combination aims to both reactivate latent HIV ("kick") and enhance immune-mediated clearance ("kill"), potentially enabling sustained viral control after discontinuation of antiretroviral therapy without significant reduction in the viral reservoir[1][2][5][6][9].
