Overview

This is a combination regimen consisting of four agents: - **N-803** (also known as ALT-803 or nogapendekin alfa inbakicept-pmln; brand name Anktiva) is a first-in-class interleukin 15 receptor agonist and IL-15 superagonist. It stimulates the proliferation and activation of natural killer cells and CD8+ T cells to enhance anti-tumor immunity. Developed by ImmunityBio, it is approved for BCG-unresponsive non-muscle invasive bladder cancer[1][2][4][5][8]. - **Carboplatin** is a platinum-based chemotherapy agent that causes DNA crosslinking and apoptosis in rapidly dividing tumor cells. - **Nab-paclitaxel** (nanoparticle albumin-bound paclitaxel; brand name Abraxane) is a microtubule inhibitor formulated with albumin nanoparticles to improve delivery. - **Pembrolizumab** (brand name Keytruda) is an immune checkpoint inhibitor targeting PD‑1 on T-cells, blocking its interaction with PD-L1/PD-L2 to restore anti-tumor immune responses. This combination leverages both direct cytotoxic effects from chemotherapy agents and enhanced immune-mediated tumor cell killing via immunomodulation. The regimen may be under investigation for solid tumors refractory to standard therapies or where synergistic immuno-oncology effects are desired[6].
