Drug intelligence / Profile preview
This is a **combination therapy** consisting of four agents: - **N-803** (also known as ALT-803) is an **IL-15 receptor superagonist complex** that stimulates and expands natural killer (NK) cells and CD8+ T cells, designed to enhance anti-tumor immunity. - **Carboplatin** is a platinum-based chemotherapeutic agent that induces DNA cross-linking and apoptosis, commonly used in lung and other cancers. - **Pembrolizumab** (Keytruda) is a monoclonal antibody that targets and blocks the **programmed cell death protein 1 (PD-1)** receptor, thereby unleashing T-cell mediated cytotoxicity against tumor cells. - **Pemetrexed** (Alimta) is an antifolate chemotherapeutic agent that inhibits several enzymes involved in nucleotide synthesis, leading to impaired DNA and RNA synthesis in cancer cells. The combination aims to synergize: N-803 activates cellular immunity to enhance the effect of immune checkpoint blockade by pembrolizumab, while carboplatin and pemetrexed act as cytotoxic agents and may provide additional immunomodulating effects. The regimen is being investigated in clinical trials primarily for advanced non-small cell lung cancer (NSCLC) and other solid tumors.
