N-803 + pembrolizumab is a combination immunotherapy regimen under investigation for the treatment of various cancers, including advanced non-small cell lung cancer (NSCLC), gastric cancer, head and neck cancer, and endometrial cancer. N-803 (also known as ALT-803 or Anktiva) is an interleukin 15 (IL‑15) receptor superagonist complex that stimulates the proliferation and activation of natural killer (NK) cells and CD8+ T cells without expanding regulatory T cells. Pembrolizumab is a monoclonal antibody targeting programmed cell death protein 1 (PD‑1), blocking its interaction with PD-L1/PD-L2 to enhance anti-tumor immune responses. The combination aims to synergistically boost immune-mediated tumor cell killing by both activating effector lymphocytes via IL‑15 signaling and relieving checkpoint inhibition through PD‑1 blockade. This regimen is being developed primarily for patients with advanced or refractory cancers who have limited options after standard therapies[2][3][5][7].
