This is a combination immunotherapy regimen consisting of three agents: **N-803** is an interleukin 15 receptor alpha fusion protein complex, also known as an IL15 superagonist, that stimulates the proliferation and activation of natural killer (NK) cells and CD8+ T cells without expanding regulatory T cells. **Pembrolizumab** is a monoclonal antibody targeting programmed cell death protein 1 (**PD‑1**) on immune cells, blocking its interaction with ligands to enhance anti-tumor immunity. **PD-L1 t-haNK** refers to allogeneic high-affinity natural killer (**haNK**) cells engineered with a chimeric antigen receptor (**CAR**) targeting programmed death-ligand 1 (**PD‑L1**) on tumor cells, enabling direct recognition and killing of cancerous targets expressing this checkpoint molecule. The combination aims to synergistically activate the immune system against tumors by enhancing NK cell activity, blocking inhibitory signals in T-cells, and directly targeting tumor-expressed PD‑L1. This regimen has been studied in advanced solid tumors including head and neck squamous cell carcinoma, gastric cancer, pancreatic cancer, non-small cell lung cancer, and others[2][4][5][8].
