Overview

N-acetyl cysteine + bromhexine is a fixed-dose combination of two small molecule mucolytic agents used primarily for the treatment of respiratory conditions characterized by excessive or thick mucus production. N-acetyl cysteine (NAC) acts as a mucolytic and antioxidant by breaking disulfide bonds in mucus glycoproteins, thereby reducing viscosity and facilitating expectoration. It also replenishes intracellular glutathione levels, providing antioxidant effects. Bromhexine is a mucolytic that enhances the production of serous mucus in the respiratory tract and depolymerizes mucopolysaccharide fibers in sputum, further decreasing its viscosity and promoting clearance from airways. Both agents have been studied individually and in combination for their potential to reduce symptoms, hospitalization rates, duration of illness, and mortality in patients with acute respiratory infections such as COVID-19[1][3][5][6]. The combination has demonstrated efficacy in clinical trials for mild to moderate COVID-19 by improving oxygenation parameters and reducing inflammatory markers[3][5]. Both drugs are well-established therapies for chronic bronchitis, COPD, pneumonia, asthma (with caution), sinusitis, rhinosinusitis, and other conditions involving mucus hypersecretion[4][7].
