Overview

[**N-methyl-[^11C]-diaminostilbene**] ([^11C]MeDAS) is a radiolabeled stilbene derivative used as a positron emission tomography (PET) imaging agent for quantifying myelin density in the central nervous system. It binds specifically to intact myelin basic protein (MBP), a key component of healthy myelin sheaths. Upon demyelination, MBP loses its structure and [^11C]MeDAS binding decreases, making it useful for detecting demyelination and remyelination processes in diseases such as multiple sclerosis. Compared to other PET tracers like [^11C]PiB, [^11C]MeDAS demonstrates higher specificity for intact myelin and can distinguish between white matter and gray matter in the brain. It is primarily used in research settings for imaging myelination status in animal models and has been evaluated in first-in-human studies[2][5][6][9].
