N-naphthalen-1-ylmethyl-2'-[3,5-dimethoxybenzamido]-2'-deoxy-adenosine is an experimental small molecule inhibitor designed as a purine 2’-deoxyribonucleoside analog. It features modifications at the adenine and ribose moieties to enhance selectivity and binding affinity for trypanosomatid glyceraldehyde 3-phosphate dehydrogenase (GAPDH), an enzyme essential for glycolysis in parasites such as Leishmania mexicana and Trypanosoma species. The compound was developed through structure-based drug design to exploit differences between parasite and human GAPDH enzymes. Structural studies have shown that it binds with high affinity to the active site of Leishmania mexicana GAPDH, inducing conformational changes that inhibit enzymatic activity and block parasite growth. There are no approved indications or clinical trials reported for this compound; it remains at the experimental stage as a potential antiparasitic agent targeting glycolytic pathways in protozoan pathogens.[1][6][10]
