Na-APR-1 (M74) + Alhydrogel + GLA-AF is a hookworm vaccine candidate that combines a recombinant version of the Necator americanus aspartic protease-1 (Na-APR-1) with aluminum hydroxide adjuvant (Alhydrogel) and an aqueous formulation of Glucopyranosyl Lipid A (GLA-AF), a Toll-like receptor 4 (TLR4) agonist. The vaccine works by inducing neutralizing antibodies that block the enzymatic activity of Na-APR-1, a hemoglobinase that facilitates the ability of adult hookworms to parasitize the human intestine and feed on blood. The Na-APR-1 component is a catalytically inactive version of the wild-type hemoglobinase (M74 mutation) that is expressed in Nicotiana benthamiana tobacco plants using an Agrobacterium tumefaciens strain engineered to express the vaccine antigen. The addition of GLA-AF to Na-APR-1(M74)/Alhydrogel has been shown to result in higher levels of IgG antibodies against Na-APR-1(M74) compared to formulations without GLA-AF.
