nab-paclitaxel + capecitabine is a combination chemotherapy regimen used primarily for the treatment of advanced or metastatic solid tumors, including pancreatic cancer and biliary tract cancers. Nab-paclitaxel (albumin-bound paclitaxel) is a microtubule inhibitor that disrupts cell division by stabilizing microtubules and preventing their depolymerization. The albumin-bound formulation enhances drug delivery to tumor tissue via gp60 receptor-mediated transcytosis and SPARC binding. Capecitabine is an oral prodrug of 5-fluorouracil (5-FU), which inhibits thymidylate synthase, thereby interfering with DNA synthesis in rapidly dividing cells. This combination has demonstrated substantial antitumor activity as first-line or second-line therapy in metastatic pancreatic cancer and advanced biliary tract cancers, with manageable toxicity profiles[1][5][8]. Nab-paclitaxel was developed by Celgene Corporation; capecitabine was developed by Roche.
