nab-paclitaxel + carboplatin + gemcitabine is a combination chemotherapy regimen evaluated as a neoadjuvant treatment for patients with locally advanced, muscle-invasive bladder cancer. The regimen includes ABI-007 (nab-paclitaxel), which is an albumin-bound nanoparticle formulation of paclitaxel designed to enhance drug delivery to tumors via albumin receptor-mediated transport while avoiding the toxicities associated with conventional paclitaxel solvents like Cremophor EL. This is combined with carboplatin, a platinum-based antineoplastic agent that induces DNA cross-linking, and gemcitabine, a nucleoside analog that inhibits DNA synthesis. The University of Michigan Rogel Cancer Center conducted a Phase 2 trial to assess the safety and efficacy of this triplet combination prior to radical cystectomy in patients whose cancer has invaded the bladder muscle wall.
