AP + Toripalimab is an investigational chemo-immunotherapy combination regimen being evaluated as induction therapy for patients with resectable locally advanced head and neck squamous cell carcinoma (HNSCC). The regimen consists of **toripalimab**, a humanized IgG4 monoclonal antibody that targets the Programmed Cell Death Protein-1 (PD-1) receptor, and the **AP chemotherapy regimen**, which comprises **nab-paclitaxel** (albumin-bound paclitaxel) and **cisplatin**. Toripalimab acts as an immune checkpoint inhibitor by blocking the PD-1/PD-L1 pathway, thereby restoring the anti-tumor activity of exhausted T-cells. Nab-paclitaxel is a microtubule inhibitor that causes cell cycle arrest, while cisplatin is a platinum-based alkylating agent that induces DNA cross-linking and apoptosis. This combination aims to enhance pathological response rates and improve surgical outcomes in the neoadjuvant setting.
