Overview
Nab-paclitaxel + FOLFOX is an investigational combination chemotherapy regimen being evaluated for the perioperative treatment of patients with operable esophagogastric adenocarcinoma. This regimen, specifically studied in the FOXAGAST phase II trial by the GERCOR Multidisciplinary Oncology Cooperative Group, combines nanoparticle albumin-bound (nab) paclitaxel with the FOLFOX backbone (oxaliplatin, 5-fluorouracil, and leucovorin). Nab-paclitaxel utilizes albumin as a delivery vehicle for paclitaxel, which targets and stabilizes microtubules to disrupt cell division. Oxaliplatin acts as a platinum-based DNA alkylating agent, while 5-fluorouracil is an antimetabolite that inhibits thymidylate synthase, and leucovorin is used to enhance the activity of 5-fluorouracil. The combination aims to improve pathological complete response rates in the neoadjuvant setting for gastroesophageal cancers.
