Overview

nab-Paclitaxel + panitumumab is a combination regimen of two antineoplastic agents used in oncology clinical trials, particularly for HER2-negative and triple-negative breast cancer. nab-Paclitaxel is an albumin-bound formulation of paclitaxel, a microtubule inhibitor that disrupts cell division by stabilizing microtubules and preventing their depolymerization. This formulation leverages albumin-mediated transport to enhance drug delivery to tumors via the gp60/caveolin-1/caveolae/SPARC pathway, increasing intra-tumoral concentrations and reducing toxicity compared to conventional paclitaxel[2][8]. Panitumumab is a fully human IgG2 monoclonal antibody targeting the epidermal growth factor receptor (EGFR). It binds specifically to EGFR on tumor cells, blocking ligand binding and subsequent receptor activation, thereby inhibiting downstream signaling pathways involved in cell proliferation, survival, angiogenesis, and metastasis[2][9]. The combination has been studied as part of neoadjuvant chemotherapy regimens for primary HER2-negative inflammatory breast cancer (IBC) and triple-negative breast cancer (TNBC), showing high pathological complete response rates in these populations[1][10].
