Overview

This is a combination regimen consisting of three agents: - **Nab-paclitaxel** (nanoparticle albumin-bound paclitaxel), a microtubule-stabilizing chemotherapeutic agent. - **S-1**, an oral fluoropyrimidine derivative combining tegafur (a prodrug of 5-fluorouracil), gimeracil, and oteracil potassium. - **PD-1 inhibitor**, a class of immune checkpoint inhibitors that block the programmed cell death protein 1 (PD‑1) pathway to enhance T-cell mediated antitumor immunity. Commonly used agents in this context include pembrolizumab, nivolumab, sintilimab, toripalimab, tislelizumab, and camrelizumab. The combination is under investigation as first-line therapy for advanced cancers such as biliary tract cancer (BTC) and gastric cancer. The rationale is to combine cytotoxic chemotherapy with immunotherapy to improve progression-free survival and response rates compared to chemotherapy alone. Early studies suggest improved outcomes with manageable safety profiles[1][5].
